FORMULATION AND EVALUATION OF BOSENTAN GASTRO RETENTIVE DRUG DELIVERY SYSTEM

ABSTRACT
The purpose of this research is to develop a novel gastro retentive tablet of Bosentan. it is an endothelin receptor antagonist effective in the treatment of pulmonary hypertension. Bosentan has short half-life (5hrs) and it has a gastric absorption. The preparation of floating tablets of Bosentan by using different grades of HPMC and gas generating agent like sodium bicarbonate, other excipients are also blended and compressed on a rotary compressed machine. The floating tablet formulations were evaluated for physical characterizations namely hardness, friability, weight variation, drug content uniformity and buoyancy studies. In vitro drug release study was performed using united states pharmacopeia XXIIV type II dissolution test apparatus
employing paddle stirrer at 50 rpm. Dissolution medium was used as 900 ml of 0.1N HCl for 12hrs, at 37 ± 0.5 0C. Among all the formulations, formulation F10 was found better as compared to other formulations.
