Abstract

OPTIMIZATION AND EVATUATION OF CLARITHROMYCIN EXTENDED-RELEASE TABLET

ABSTRACT

Primary objective of sustained drug delivery system is to ensure safety and to improve efficacy of drug substance as well as the patient compliance which can be achieved by better control of plasma drug levels and less frequent dosing of the drug. For any sustained or extended-release dosage form it is very important to use of minimum number of excipient material with minimum processing steps in order to reduce the tablet-to-tablet variation and batch-to-batch variations, hence direct compression type is the most suitable technique of clarithromycin preparation. Clarithromycin comes as a tablet and a suspension (liquid) to take by mouth. The regular tablet and liquid are usually taken with or without food every 8 (three times a day) to 12hours (twice a day) for 7 to 14 days. But having some unwanted side effects like abdominal pain, diarrhea, dyspepsia, nausea, vomiting, flatulence, dysphagia, constipation, gastritis, stomatitis, peptic ulceration with or without bleeding. One approach to overcome the undesirable side effects of Clarithromycin drug includes the use of sustained release tablets as controlled release delivery through which the tablet dose may change from 3 times to one time every 24 hours (once a day) for 7 to 14 days.
