IDENTIFICATION AND ISOLATION OF FLAVONOL DERIVATIVE GALANGIN FROM ANISOMELES MALABARICA AND ITS EFFICACY ON APOPTOSIS AND DNA FRAGMENTATION OF HUMAN BREAST CANCER CELLS

ABSTRACT
The goals of the present investigation were to identify and isolate the bioactive compound from Indian herb Anisomeles malabarica. Leaves methanol extract was obtained using Soxhlet apparatus, purified by successive extractions with solvents i.e. hexane in chloroform, hexane in ethyl acetate, and hexane in methanol (90:10, 80:20, 70:30: 50:50, 30:70). The active fraction was separated by column chromatographic and separated by thin layer chromatography and was characterized by FTIR, 1H NMR, 13C NMR and Mass Spectrometry. The spectral study results show that, presence of flavonol (Galangin) in methanol extract (Petroleum ether: Ethyl acetate: Methanol (9:0.5:0.5). Further, GALANGIN was evaluated for its cytotoxicity on Normal cell lines and its antiproliferative efficacy against different cancer cell lines
(A549, Hep3B, HeLa & MCF-7) by MTT Assay. The results indicated that, Galangin significantly inhibit the growth of MCF-7 cell lines (21.10 μg/mL) than the other cell lines. It was found that; Galangin could significantly inhibit the growth of MCF-7 cells in a dose and time dependent manner and it also inhibits the cell proliferation and early apoptosis through G0-G1 phase and down regulation of antiapoptotic protein expression by regulating the caspase signalling cascade. Therefore, Galangin has the potential to be developed as an anti-cancer agent against breast cancer after clinical trials.
