Abstract

DEVELOPMENT AND EVALUATION OF POLYURETHANE FOAM BASED DRESSING SYSTEM FOR DIABETIC WOUND

ABSTRACT

Background: The present work is based on development of foam based dressing system for diabetic wound containing asiaticoside and hyaluronic acid. Diabetic wound has different pathological conditions than normal wound. Wound healing mechanism is aggreviated due to impairment in angiogenesis, failure in fibroblast, keratinocyte, matrix metalloproteinase (MMP) activity, irregular growth factor activation and increased oxidative stress. Therefore, a multifaceted approach is required for diabetic wound healing. Asiaticoside is triterpene glycoside in nature isolated from centella asiatica which increases collagen synthesis, promotes angiogenesis and Hyaluronic acid is natural component present in Extracellular matrix having anti-inflammatory as well as anti-bacterial properties. Hyaluronic acid promotes growth factor activation, fibroblast proliferation and reduce oxidative stress which aids in wound healing process. Materials and Methods: Initially asiaticoside and hyaluronic acid is mixed with aloe vera juice. Carbopol 934 is used as gelling agent along with other ingredients. This gel is incorporated into sterile polyurethane foam dressing. The dressing system is checked for in vitro and in vivo activity. Results: The dressing system shows good homogeneity, grittiness and folding endurance. The incorporated gel has good antibacterial activity against staphylococcus aureus and E. coli. In vitro diffusion was reported to be 93.67 % for asiaticoside and 80.67 % for hyaluronic acid. Alloxan induced diabetic rats are used for wound healing activity in excision wound model. Complete wound closure is observed at the end of day 15.
