Abstract

STANDARIZATION OF GINGER RHIZOME EXTRACT AND DETERMINATION OF [6]-GINGEROL CONCENTRATION IN THE ETHYL ACETATE FRACTION BY TLC - DENSITOMETRY

ABSTRACT

Ginger rhizome is widely used as an ingredient of traditional medicine in Indonesia. Ginger rhizome (Zingiber officinale Roscoe) shown an anti-cancer activity, chemical compound responsible for anticancer is [6]-gingerol. To obtain [6]-Gingerol from ginger rhizome, the active substance used in this study was the ethyl acetate fraction of ginger rhizome (Zingiber officinale Rosc.). The purpose of this study was to develop a quantitative analytical method for [6]-gingerol in ginger rhizome fraction using the TLC-Densitometry method. The results of the standardization of the extracts were organoleptically in the form of thick extracts, brownish yellow color, with the aromatic odour of ginger. The yield of the extract was 12.2%, drying shrinkage of 4.55% ± 0.004%, the total ash content of the ethanol extract was 0.03 ± 0.01%. Determination of the levels of [6]-gingerol using the TLC-Densitometry method which has been validated including linearity, obtained the linear regression equation y = 10.18x + 1055.1. The detection limit of 6-gingerol was 71.92 g/mL and the quantitation limit was 239.7 g/mL. The coefficient of variation value is 1.7%. So that the level of [6]-gingerol in the ethyl acetate fraction of ginger rhizome is 6.118%.
