STOMACH SPECIFIC FLOATING CUM MICOADESIVE DRUG DELIVERY OF OSELTAMIVIR MICROSPHERES TO TREAT VIRAL DISEASES

ABSTRACT
The goal of any drug delivery system is to provide a therapeutic amount of drug to the proper site in the body to achieve promptly & then maintain the desired drug concentration. The present research work was an attempt to develop and evaluate stomach specific microparticles for anti-influenza drug as Oseltamivir phosphate. This drug was recommended by WHO to treat flu symptoms caused by influenza virus. Floating cum mucoadhesive microspheres were prepared by emulsification heat denaturation method using different polymers in order to increase the release by improve buoyancy and mucoadhesion. Nine various formulations of Floating Mucoadhesive (FM) Microspheres (F1-F9) were developed with different ratios of chitosan and albumin combinations. The Fourier transform infrared (FTIR) spectroscopy was used to confirm compatibility and to rule out any possible interactions between drug and polymers. All formulations were prepared by heat denaturation method using gelatin as gelling agent, sodium bicarbonate as stabilizer, sorbitan as emulsifying agent, liquid paraffin as stiffening agent. Physicochemical characterisation in terms of particle size, percentage yield, drug entrapment efficiency, Swelling study, buoyancy, mucoadhesion, in vitro drug release of all formulations were found to be within acceptable range. Among all the prepared formulations, F7 was found to be an optimised formulation with optimized drug release and it was found to be best formulation. This may treat flu symptoms also be given to prevent influenza in people who may be exposed but do not yet have symptoms of corona.
