Abstract

EVALUATION OF ANTICANCER EFFECT OF COLEUS MALABARICUS BENTH. AND ITS MORPHOTYPE ON SK-MEL-28 CELL LINE

ABSTRACT

Cancer is still a growing health problem in both developing and developed countries. Phytochemicals are increasingly used for the prevention and treatment of various malignancies owing to the ease of availability with reduced side effects. The potential therapeutic importance of the family Lamiaceae is related to volatile oil components and flavonoids. The aim of the study was the investigation of cytotoxic activity of alcoholic extract of Coleus malabaricus Benth. and its morphotype by invitro MTT assay on SK-MEL-28 human melanoma cell lines. The study is based on the objective is that even though a morphotype, the second plant might have some differences in the chemo taxonomical point of view. The phytochemical analysis showed the presence of alkaloids, phenolic compounds and tannins, flavonoids, terpenoids and carbohydrates in the plant extracts. Cytotoxicity findings were based on cell viability and calculation of IC50 values. The results indicated that in the range of tested concentration used (6.25g/ml to 50 g/ml) there is a decrease in the optical density values as well as cell viability. The most significant reduction was noticed at a concentration of 50 g/ml. IC50 values for Coleus malabaricus, morphotype and 5-Fluorouracil are 36 g/ml, 32g/ml and 13g/ml respectively. Results showed that %viability of cells is directly correlated with increasing concentrations of plant extracts and it is significant(p value < 0.0001) when compared with 5-Fluorouracil .Findings of the study proved that like other Coleus species, Coleus malabaricus is also would be a promising candidate for the prevention and treatment of this dreadful disease.
