FORMULATION AND IN-VITRO EVALUATION OF K-CARRAGEENAN NANOGEL OF METFORMIN DRUG

ABSTRACT
The main objective of the present work was to formulation and invitro evaluation of nanogel of metformin. Metformin is an FDA approved and well accepted antidiabetic drug which has antagonistic effect on pancreatic cancer proliferation. Moreover, decreased growth of pancreatic cancer xenografts in nude mice models administered with MET emphasizes its ability as a pancreatic cancer drug. The major problems related to MET are its low bioavailability and short half life period. Thus, in order to overcome these problems, we have developed chitosan based nano composites for controlled MET antidiabetic drug
Delivery. An alternative approach to control release involves use of nanogel employing hydrophilic natural hydrogel polysaccharide like k-carrageenan. This polysaccharide is use because it is biodegradable, nontoxic, renewable, abundant in nature and relatively cheap as compare to synthetic polymer. In the present investigation, a nanogel delivery system of metformin Hydrochloride capable of providing controlled release was prepared using different concentration of kappa carrageenan as well as different concetration of potassium chloride. The obtained nanogel after lyophilization free flowing. A higher encapsulation efficiency of drug and percentage yield was obtained in formulations A7 among all formulation that was 94.58±0.061 and 94.37±0.069 due to Drug-Polymer ratio. The Particle size of formulation A7 was 410.8nm and PDI 0.381 while zeta potential of formulation was found to be -17.9 mv. The drug release from the nanogel prepared in formulation A7 achieved its target i.e. 96.29±0.091. According to model fitting methods the highest regression coefficient (R2) value was 0.9839 through Higuchi model.
