ABSTRACT
Recently we have noted in vitro antioxidant activity of methanol extract of Calamus rotang (C. rotang) leaves. As pharmacological activity of a plant depends on temperature and time of extraction process, aim of the present work was to see effects of temperature and time of the extraction process on in vitro antioxidant activity of C. rotang leaves. C. rotang leaves were collected and identified by the taxonomist. Methanol extracts of the leaves were prepared separately maintaining different temperatures and time. In vitro antioxidant activity of each extract was measured through superoxide anion generation by xanthine-/xanthine oxidase assay, linoleic acid peroxidation assay and by DPPH photometric assay. Quercetin was used as standard antioxidant. Results showed that methanol extract of
C. rotang leaves extracted for 10 minutes at 600 C showed maximum antioxidant activity which was comparable to that of quercetin. These conditions may be maintained during isolation work of the antioxidant compound from C. rotang leaves.
Keywords: Calamus rotang Leaves, effect of temperature, effect of time, xanthine-/xanthine oxidase assay, linoleic acid peroxidation assay, DPPH photometric assay.
