PELLETIZATION A PROMISING TECHNIQUE FOR SUSTAINED DRUG DELIVERY SYSTEM

ABSTRACT
Sustained release pellets release of drug continuously in predetermined pattern over an extended period of time. They have many therapeutic advantages compared to single unit dosage forms in drug absorption, distribution, patient compliance, safety, efficacy and they release drug at regular time interval & at right site of action to maintain therapeutic range of drug in blood plasma. Sustain release systems are a better option for drugs with short half-lives that require repeated dosing since they are simple to formulate and independent of the gastrointestinal tract's absorption process after oral administration. The present review outlines the manufacturing of sustained release pellets. Sustained pellets prepared by a number of techniques, include layering, cryopelletization, freeze pelletization, spray drying, spray congealing,
extrusion, spheronization and hot melt extrusion have been discussed. The basic information about sustained-release formulation, its advantages and disadvantages, drug selection for sustained release, and factors involved in oral sustained-release dosage form design are all covered in this review article.
