DEVELOPMENT AND VALIDATION OF RP-HPLC METHOD FOR THE DETERMINATION OF SULFISOXAZOLE IN RAT PLASMA

ABSTRACT
A Simple, Precise, Accurate and rapid liquid chromatography (RP-HPLC) method has been developed and validated for the determination of Sulfisoxazole in rat plasma. A reverse phase Eclipse plus C18 column (250mmX4.6mm,3μm) with mobile phase consisting of sodium acetate buffer (pH 4.0 adjusted with glacial acetic acid): acetonitrile in the ratio of 60:40%, v/v at the flow rate of 1 ml/min and UV detection wavelength at 266 nm with a total run time of 10 min. The retention time for Sulfisoxazole was found to be 4.9/min. The method was linear over the concentration range of 25-150 μg/ml. The percentage mean recovery for Sulfisoxazole was observed to be 99.3% to 102% and the RSD was observed under 1%. The method was successfully demonstrated for the determination of Sulfisoxazole in rat plasma and validated as per ICH guidelines and the results were found to be within the limits. The proposed method was a new method of analysis for assessment of Sulfisoxazole in rat plasma by RP-HPLC method.
