Abstract

A REVIEW ARTICLE ON SYNTHESIS, ISOLATION AND CHARACTERIZATION OF DONEPEZIL HYDROCHLORIDE IMPURITY

ABSTRACT

Donepezil, sold because the trade name Aricept among others, could be a medication accustomed treat Alzheimer's disease. It appears to lead to atiny low benefit in mental function and skill to function. Use, however, has not been shown to vary the progression of the disease. Treatment should be stop if no benefit is seen. it's taken orally. Donepezil binds reversibly to acetylcholinesterase and inhibits the hydrolysis of acetylcholine, thus increasing the supply of acetylcholine at the synapses, enhancing cholinergic transmission. Donepezil is slowly absorbed from the Gastrointestinal tract. Its maximal plasma concentrations were reached within 3-4 hours. Its relative oral bioavailability is 100% and food failed to affect its absorption. Pharmacokinetics are linear over a dose range of 1–10 mg given once a day. Neither food nor time of administration influences the speed and extent of its absorption. it's approximately 96% guaranteed to plasma proteins. The drug is both excreted by the kidney and extensively metabolized by the liver CYP 450 (2D6 and 3A4) still as glucuronidation. The elimination half-life is about 50–70 hours and also the mean apparent plasma clearance (Cl/F) is 0.13 L/hr/kg. Common side effects include nausea, trouble sleeping, aggression, diarrhea, feeling tired, and muscle cramps.Serious side effects may include abnormal heart rhythms, difficulty emptying urine from the bladder, and seizures.
