THE CONTENT DETERMINATION OF AMPLEXICOSIDE G IN MERRILLANTHUS HAINANENSIS

ABSTRACT
In order to develop a method for the quality assessment of Merrillanthus Hainanensis Chun Et Tsiang, the content of Amplexicoside G (1) was determined by HPLC. The chromatographic conditions were as follow, column: Phenomenex Luna C18 (250 mm×4.6 mm, 5 μm); mobile phase: acetonitrile (A)-0.1% formic acid water (B); elution gradient: 10 to 50% A in 30 min, 50 to 65% A in 5 min, 65 to 100% A in 20 min and 100% A in 10 min; flow rate: 1.0 mL/min, column temperature: 30℃, and detection wavelength 296 nm. The results showed that compound 1 exhibited linear relationship in the range of 0.086~1.49 g (r = 0.9995), and the average recovery rate was 98.20% with RSD of 1.07%. The content of compound 1 from 12 batches of M. Hainanensis was in the range of 4.31~14.79 μg/mL. The content of compound 1 in different batches exhibited significant variation due to different collection time. Our developed method was rapid, simple, reproducible, stable and reliable, and could be used for the content determination of compound 1 in M. Hainanensis.
