Abstract

FORMULATION AND EVALUATION OF SUSTAINED RELEASE MUCOADHESIVE MICROCPAASULES OF BACLOFEN

ABSTRACT

Baclofen is muscle relaxant drug used for the treatment of spastic movement disorder, especially in instances of spinal cord injury, multiple sclerosis and also cerebral palsy. Spasm generally result for several hours to minutes. When baclofen is administered orally as a tablet, it is rapidly and completely absorbed and hence increasing the dosing frequency. In the present study an attempt is made to formulate sustained release dosage form of baclofen using chitosan, Carbopol 934P and HPMC K4M as release retarding polymers to reduce dosing frequency and improve patient compliance. Also HPMC and carbopol were used as mucoadhesive polymers. Various batches of microcapsules were developed by varying concentration of chitosan, carbopol and HPMC K4M by using ionic gelation method. Developed mucoadhesive microcapsules were evaluated for various parameters like Bulk density, Tapped density, carr’s index, Hausner’s ratio, Percentage yield, Drug entraptment efficiency, mucoadhesive strength, In-vitro drug release study etc. The formulation F6 found to be optimum based on all evaluation parameters. Also the optimized formulation F6 found to be stable for one month and dosen’t change its colour, shape, release pattern.
