THYMOQUINONE PLGA-VITAMIN-E TPGS NANOPARTICLES FABRICATION: A RATIONAL DESIGN THROUGH BOX-BEHNKEN DESIGN

ABSTRACT
Thymoquinone (TQ) a vital phytoconstituents from essential oil of Nigella sativa is long-established for possessing therapeutic advantages in several ailments. Its Lipophilic and hard-hitting plasma protein-binding tends transitory bioavailability and swift elimination, to prove any substantial potential. Current research intended to offer an effective design for Thymoquinone nano-formulation, for a prospective treatment approach. Independent variable’s influence viz PLGA conc. (X1), Vitamin E-TPGS %w/v (X2) and ultra-sonication time (X3) on dependent Responses viz Particle size (R1), %EE (R2) and Cumulative Release (R3) were optimized, employing Box-Behnken approach. Optimized and predicted independent variables were generated as 144 mg PLGA with 2.03% Vitamin-E-TPGS and 12.24 seconds for ultrasonication time. Generated dependent variables viz particle-size of 195.5 nm, %EE of 73.4 and Cumulative release of 59.1%. TQN was optimized via Box-Behnken Design, and projected as Thymoquinone nano-delivery imparting benefit towards variety of ailments.
