Abstract

PHARMACOLOGICAL INVESTIGATION FOR ANALGESIC AND ANTI-INFLAMMATORY ACTIVITY OF HERBAL EXTRACTS USING ANIMAL MODELS

ABSTRACT

Ficus dalhousiaemiq (Fd) and Annona muricata (Am) are utilized as herbal medicines. Fd isemployed traditionally as anti-septic, haemostatic, skin diseases, dysentery, diarrhea, ulcers, deficient lactation, leucorrhoea, menorrhagia and veginal disorders while Am is employed as diabetic, anti-hypertensive, anti-bacterial and parasitic infections, fever, respiratory and skin illnesses. Theethanol extracts of ficusdalhousiae and Annona muricata can be utilized as potent analgesic and anti inflammatory drugs as they had exhibited significant effects against pain and inflammation. The extraction process of extricates was obtained through soxhlet technique resulting Ethanol extracts of plant in short span, reveals the phyto chemical constituents as alkaloids, flavonoids, phenolic compounds, amino acids, carbohydrates, sterols and saponins. Aspirin was utilized as standard drug for pain 20mg/kg dose while indomethacin for inflammation at 10mg/kg dose. FDEE and AMEE were utilized at 200mg/kg and 400mg/kg singly and revealed more significant when used in combination i.e FDEE+AMEE at 400mg/kg. Explorations were made with two analgesic models in rat namely Eddy‟s hot plate test and Acetic acid induced writhing test while anti-inflammatory models were accomplished through Carrageenan induced paw edema model and Egg albumin induced paw edema model. These analgesic and anti-inflammatory exploratory results disclose that FDEE and AMEE in combination at 400mg/kg significantly reduced the licking response time during hot plate test and writhing counts during writhing test. In addition, notable reduction in paw volume was observed both in carrageenan and Egg albumin induced inflammation. The invitro methods were evaluated and ic50 values of naloxone assay and cox assay was compared to standard in both cases ficusdalhousiae showed effectiveness compared to annona muricata. The parameters such as histamine, PGE2, interleukins 8, 6, and 1, cytokines were evaluated and Fd+Am showed significance in suppressing the raised levels of the mediators. The present investigations revealed the analgesic and anti-inflammatory effects of FDEE and AMEE substantiate potent traditional medicines.
