ABSTRACT

The purpose of this study was to prepare and characterize microspheres loaded by Topiramate. To achieve this goal Chitosan and STPP microsphere loaded by Topiramate were prepared by Ionotropic gelation method. The use of Ionotropic gelation method avoids the possibility of the occurrence of the toxic and undesirable effects associated with the use of glutaraldehyde, a chemical cross linking agent. Microspheres are characteristically free flowing powders having particle size ranging from 1-1000 μm consisting of proteins or synthetic polymers. Morphology, size, encapsulation efficiency and drug release from these microspheres were evaluated. Microscopic evaluation of microsphere showed that microspheres were spherical in shape. The size analysis result indicated that size range varied from 223.44 to 246.96 μm. Encapsulation efficiency of microspheres was increased by increasing drug to polymer ratio. FTIR spectroscopic analysis was performed to ascertain drug polymer interaction, if any during the microsphere preparation. The surface morphology of the prepared microsphere was studied by SEM. From the results of the present investigation it may be concluded that drug loaded chitosan microspheres can be prepared by a simple technique which avoids the use of complex apparatus and special precaution.
