SCREENING AND CHARACTERIZATION OF KETOCONAZOLE CO-CRYSTAL USING SOLVENT ASSISTED GRINDING APPROACH FOR SOLUBILITY ENHANCEMENT

ABSTRACT
The present study attempt for the formulation and evaluation of co-crystal of poorly water soluble drug. Pharmaceutical co-crystal is emerging method for altering the physicochemical properties of pharmaceutically active ingredient such as solubility and dissolution rate. Ketoconazole is a BCS class II antifungal drug with poor water solubility, this co-crystal approach used to formulate co-crystal with ascorbic acid Coformer by using Solvent assisted grinding method. The prepared co-crystal formulation was characterized and evaluated by Fourier transform Infrared spectrophotometer, Melting point determination, Solubility, Drug content, percent practical yield, flow property, and In-vitro Dissolution study. Based on this study,
concluded that the co- crystallization of ketoconazole with ascorbic acid offers a valuable improvement in the the present study, it found that formulation F1 and F2 show in vitro drug release 82.89% and 89.90% respectively within 90 mins. solubility, Dissolution rate, and flow properties as compared to pure drug.
