SILYMARIN MICROSPHERES AND ITS ANTIPROLIFERATIVE ACTION ON HUMAN PROSTATE ADENOMATOUS CANCER DU145 CELLS

ABSTRACT
Silymarin a flavonoid from milk thistle(Silybum marianum) exhibits a variety of pharmacological actions, including anti-proliferative and apoptotic activities against various types of cancers in intact animals and cancer cell lines. This present study was aimed to formulate microspheric form of Silymarin by using various polymers like Chitosen, Pectin, Carbopol 934P, Ethyl cellulose, Eudragit RS100. Microspheres were successfully prepared by emulsion solvent evaporation and ionic cross linking method.The prepared microspheres were evaluated for various parameters like encapsulation efficacy,percentage yield, invitro release and evaluated for their invitro cytotoxicity potential. The obtained yields varied from 46-91% on various polymers and encapsulation efficacy was up to 91.12%. Results from In vitro release studies indicated that, Ethylcellulose and EudragitRS100 microspheres exhibited prolonged drug release for 12h, and, therefore, could potentially improve the bioavailability of the Silymarin. Results from invitro cytotoxic studies showed that,the maximum percentage of cell viability is reduced up to 25 Percentage at 12hour which clearly indicates that 75 percentage cell deaths occurred in DU145 Prostate carcinoma cells. DNA Fragmentation study also confirmed better better anti cancer activity of Silymarin microspheres against human prostate adenocarcinoma cellline DU145.Hence the formulations can be effectively tested for its anticancer activity.
