ABSTRACT
Introduction – Ayurveda has many wonder formulations with greater clinical result. But for the global acceptance and scientific validation Analytical study is important in validating the formulation. Gritha Kalpana is a unique preparation where Gritha takes the quality of drugs used and make it more potent.[1] Panchagavya Gritha is the preparation prepared by using only Panchagavya and used in Manasika Vyadhi Apasmara and Sharirika Vyadhi Jwara. According to Acharya Charaka ratio of Panchagavya in Panchagavya Gritha[2] is equal but Acharya Vagbhata[3] did not mention about ratio of drugs used in the preparation. Materials and method – Pancha Gavya is collected freshly and test done for adulteration for Ghee, milk and Dadhi. Panchagavya
Gritha is prepared in two batches at the practical laboratory of Muniyal institute of Ayurveda medical science, Manipal, preparation with all drugs in equal ratio is named as PGG1 and with drugs in Anukta[4] ratio is named as PGG2. Result – LOD[5] was well within the normal limit in both samples. Specific gravity[5] is higher in PGG2 and PGG 2 showed rancidity after 3 months of preparation where as PGG1 samples were stable. All the samples showed higher iodine value[5] compared to the raw ghee. Saponification value was decreased in PGG1 and increased in PGG2 as time advanced. In HPTLC[6] analysis variable components from 3 to 6 were found. Discussion and conclusion - As Ghrita contains essential fatty acids that also act as vehicles for fat soluble vitamin is considered as superior among Sneha Dravya. Panchagavya Ghrita with five components has been recommended for the treatment of Apasmara, Jwara and Kamala in classics. HPTLC study was found to be useful parameter for the as the separation was satisfactory. But picks couldn’t be characterised due to lack of markers, however in total more active compounds were observed in PGG1 samples. When considered with the parameters like less time duration, more stability and active compounds in HPTLC ratio used in PGG1 sample seems to be ideal.
