INSIGHTS INTO SUSTAINED RELEASE DOSAGE FORMS PROSPECTS AND CHALLENGES: A REVIEW

ABSTRACT
Sustained release dosage forms modify and improve the drug performance by increasing the duration of drug action, to improve bioavailability, to increase patient compliance, to reduce the number of doses and to achieve a prolonged therapeutic effect by continuously releasing medication over an extended period of time after administration of single dose. Sustained release drug delivery system optimizes the pharmacokinetic, pharmacodynamics and biopharmaceutical properties of a drug in such a way that its side-effects are reduced, utility is maximized and cure of the disease is achieved. Matrix tablets are the oral solid dosage forms in which the drug is homogeneously dispersed or dissolved within the hydrophilic
or hydrophobic polymeric matrices. The preparation of sustained release matrix tablets involves the direct compression of blend powder mixture of drug, retardant material and other additives to formulate a tablet in which the drug is dispersed in a matrix of the retardant. There are various numbers of natural polymers which have been investigated as sustained release agent like guar gum, pectin, micro crystalline cellulose. The current review focuses on the oral sustained release tablet, their formulation, quality control, release kinetics and prospects and challenges related to material selection and tableting.
