DEVELOPMENT AND VALIDATION OF RAPID RP-HPLC-PDA METHOD FOR THE ANALYSIS OF CURCUMIN IN BULK FORM

ABSTRACT
The aim of present study was to develop a rapid, efficient and economical RP-HPLC-PDA method for the estimation of Curcumin (CUR) in bulk form. The chromatographic separation was achieved on Phenomenex kinetex C18 reverse phase column (150×4.6mm, 5μm) employing a mobile phase consisting of 0.02% formic acid: acetonitrile (45:55v/v) at a flow rate of 1.5mL/min in isocratic mode. Eluents were monitored at 425nm using PDA detector with an injection volume of 30μL. Under these LC conditions Curcumin was eluted at 2.95min and a good linearity was observed over a concentration range of 0.1-2μg/mL (R2 = 0.999). The limit of detection and limit of quantification were 0.008μg/mL and 0.024μg/mL respectively. The developed method was validated as per ICH Q2B guidelines for the parameters like accuracy, precision, linearity, specificity and robustness and the validation acceptance criteria were met in all the cases. The developed method was successfully used for the estimation of Curcumin in bulk form.
