ENHANCEMENT THE DISSOLUTION PROFILE OF CIPROFLOXACIN AND METRONIDAZOLE WITH ?-CYCLODEXTRIN AND ACIDIC SOLUBILIZING ADDITIVES

ABSTRACT
The objective of present study is improvement of solubility profile of a poorly soluble drug of ciprofloxacin and metronidazole at colonic site by incorporating some amount of solubilizing additives i.e. ascorbic acid and citric acid into the β-cyclodextrin complexes. Ciprofloxacin and metronidazole drugs having lower solubility profile in water, 0.1 N HCl, at stomach and intestinal pH. Drug containing β-cyclodextrin complexes with ascorbic acid and citric acid increased their solubility profile at different dissolution media. The solubility profile of both drugs in water showed that the inclusion complex is stable. Result
showed the solubility of drugs increased upto 100 times as the concentration of β-cyclodextrin increased. Addition of solubilizing additives such as citric acid and ascorbic acid in various proportions affect the dissolution profile of drugs in different dissolution media. The inclusion complex containing β-cyclodextrin with acidic solubilizing agent increased dissolution rate in colonic environment. This result shows increased bioavailability of both drugs at site specific environment. FTIR spectral studies performed on the solid complexes have shown that there is no interaction of the drug with the additives and β-cyclodextrin. Disc diffusion study of complex shown larger diameters of zone of inhibition and result indicated that a greater diffusivity of the drug into the agar medium.
