DEVELOPMENT AND VALIDATION OF SECOND DERIVATIVE SPECTROPHOTOMETRIC METHOD FOR ESTIMATION OF TROPICAMIDE IN PRESENCE OF CHLORBUTOL AS PRESERVATIVE

ABSTRACT
A simple and rapid second derivative spectrophotometric method for the estimation of Tropicamide in the presence of preservative Chlorbutol has been developed and validated. The Tropicamide solution was scanned in the UV region from 200-400 nm at medium speed and the spectra obtained was transformed into second derivative (Δλ = 2 nm). The amplitude was measured at 256 nm. The method was found to be linear (R2 > 0.997) in the range of 5 – 50 μg/mL for Tropicamide with detection and quantitation limits 0.902μg/mL and 2.734μg/mL respectively. Assay for Tropicamide was found 99.50% with a relative standard deviation of 0.37%. The method was effectively applied for the determination of Tropicamide in ophthalmic formulation without any interference from the excipients as well as the preservative.
