Abstract

IN-VITRO EVALUATION OF ANTIOXIDANT AND CYTOTOXIC ACTIVITY OF ETHANOLIC EXTRACT OF POUTERIA CAMPECHIANA (KUNTH)BAEHNI. BARK.

ABSTRACT

Background: Pouteria campechiana Kunth Baehni (P.C.) is a member of the family sapotaceae. P. C. is a native to Central America, and it is widely distributed around the tropical and subtropical regions in South America and Asia. The astringent decoction of the bark is used as a febrifuge in folkloric medicine. The present study was undertaken is to explore the anticancer activity of the ethanolic bark extract of Pouteria campechiana (Kunth) Baehni using in vitro models. Methods: Ethanolic extract of Pouteria campechiana (EEPC) subjected to preliminary qualitative phytochemical investigation by using standard procedures. EEPC was screened for its in vitro activity by cytotoxicity studies, determining apoptosis by using acridine orange and ethidium bromide double staining method. To establish the possible mechanism of action by Flow Cytometry and RT-PCR analysis. Results: In vitro cytotoxicity studies showed a concentration-dependent decrease in percentage viability. Treatment with EEPC resulted in apoptotic cell death and also induced significant G0/G1 phase arrest that is independent of WAF/P21 upregulation. Conclusion: We observed that the ethanolic extract of Pouteria campechiana possesses significant antitumor activity in in-vitro studies.
