FREE RADICAL SCAVENGING POTENTIAL OF LEUCOPHYLLUM FRUTESCENS THROUGH TWO OPTIMIZED SPECTROSCOPIC TECHNIQUES

ABSTRACT
Present study explains for the first time, scavenging potential of Mexican, American native medicinal plant Leucophyllum frutescens by two different techniques. i.e. UV-spectrometry and well plate method. Methanolic extract of Leucophyllum frutescens was subjected to scavenging potential, in which 1,1-diphenyl-2- picrylhydrazyl (DPPH) was used as a stable free radical. Percent scavenging potential was determined by two optimized methods and their co-relation was determined among them. IC50 value for Leucophyllum frutescens and ascorbic acid by UV-visible spectrophotometer method (B1) was determined as 318.147 μg. ml-1 and 26.157 μg. ml-1, respectively. Ratio between Leucophyllum frutescens and L-ascorbic acid by UV-visible spectrophotometer method was calculated 12.161. IC50 value for Leucophyllum frutescens and ascorbic acid by microplate reader method (D2) was determined as 761.942 μg. ml-1 and 60.946 μg. ml-1, respectively. Ratio between Leucophyllum frutescens and L-ascorbic acid by microplate reader method was calculated 12.161. Ratio of Leucophyllum frutescens and L-ascorbic acid by both methods i.e. B1 and D2, were correlated and no significant difference was found. Presented optimized methods can be successfully employed for determination of free radical scavenging activity or antioxidant activity of any plant extract or isolated compounds.
