SOLUBILITY ENHANCEMENT OF DIACEREIN BY USING SOLID DISPERSION TECHNIQUE

ABSTRACT
Diacerein, a naturally occurring molecule with numerous pharmacological activities. But due to its limited aqueous solubility and degradation at alkaline pH reduces its bioavailability. Solid dispersions of diacerein with hydrophilic polymers such as PEG 4000 and HPMC K15 M in the weight ratio of 1:2:2; 1:3:3 and 1:4:4 respectively were prepared by solvent melt method. Physical characterization with IR, DSC, SEM, FTIR, XRD studies revealed the changes in the solid state during preparation of solid dispersion and revealed the formation of high energy amorphous phase. Both solubility and dissolution rate of diacerein solid dispersions were
enhanced as compared to pure drug. It is also observed that solid dispersion of diacerein with PEG 4000 and HPMC K15 M in 1:4:4 ratio gives better dissolution as competed with other ratios.
