Abstract

ABSORPTION MAXIMA AND UV-VISIBLE SPECTROPHOTOMETRIC METHOD FOR ESTIMATION OF SORAFENIB TOSYLATE

ABSTRACT

Sorafenib tosylate, an anti cancer drug used for treatment of hepatocellular carcinoma and pancreatic cancer was approved by the U.S food and drug administration in December 2005. In the present, a simple, precise, and accurate method of estimation of sorafenib by absorption maxima method has been developed for use in formulation development, It is practically insoluble in water, solubility studies by using cosolvents indicated complete aqueous solubility using 1% v/v DMS as cosolvents, hence 100μg/ml solution of sorafenib tosylate in 1% v/v of DMSO was seamed between 200-400nm. As there is no reported simple UV spectrophotometric method for estimation of sorafenib tosylate, the aim of the present work was to develop simple, accurate and precise UV spectrophotometric method for estimation of sorafenib tosylate by absorption maxima method. 1 ml of DMSO (freely soluble) is used to dissolved the drug it is insoluble in water and diluted with water as a solvent. 30μg/ml solution of sorafenib tosylate was used as working standard solution. The solution was scanned in the wavelength range 200-400nm against the reagent blank to obtain the absorption maxima using UV method. Clear peak absorbance appeared at 261nm was considered as absorption maxima, λmax. Calibration curve was constructed at 261nm for dilutions containing 10, 20, 30, 40, 50, 60μg/ml of sorafenib tosylate. The curve was obtained ‘r’ value of indicating positive correlation between of sorafenib tosylate and corresponding absorbance values. These method obeys beer’s law in the range of 10-100μg/ml.
