APPLICATION OF RESVERATROL-LOADED CUBIC LIQUID CRYSTAL NANOPARTICLES

ABSTRACT
Based on previous studies on the preparation and characterization of resveratrol-loaded cubic liquid crystalline nanoparticles (RES-CLCNPs), in this article, we investigated the antitumor activity of RES-CLCNPs. Its effects on the proliferation of A549 and HepG2 cells were evaluated by MTT assay. With the increasing of drug concentration, the inhibition rate of RES-CLCNPs showed a certain dose-dependent. The same concentration of RES-CLCNPs acted on A549 and HepG2 cells at different times, cell inhibition rate increased when the action time extended. Under the light microscope, the number of cells is small, the diopter is low and the distribution is loose.
Fluorescent microscopy showed bright blue fluorescence, and the chromatin was condensed and the nucleus was gathered. Flow cytometry (FCM) was used to detect the impact of RES-CLCNPs on cell cycle and apoptosis, the results showed that the arrest of cell tends to G2/M phase and early and late arrest is obvious. Western-blot showed that RES-CLCNPs may be through the promotion of Bax expression and inhibition of the expression of Bcl-2 to induce apoptosis of tumor cell, and RES-CLCNPs inhibited the expression of Bcl-2 in A549 cells stronger than HepG2 cells. This article will provide a good guidance for the development of some drugs with characteristics of insoluble, short half-life and/or low bioavailability.
