Abstract

CURCUMIN LOADED CHITOSAN NANOPARTICLES FORMULATION AND OPTIMIZATION BY IN VITRO AND EX VIVO EVALUATION

ABSTRACT

Chitosan containing curcumin nanoparticles were prepared aiming at novel delivery of drugs to the site of action. The nanoparticles were prepared by using ionic gelation method. The optimization of the formulation parameters were done by using design expert software v10. A 24 full factorial design was developed by the software. Chitosan concentration, Acetic acid concentration, sodium tripolyphosphate(TPP) concentration and pH were the factors and the responses were particle size, encapsulation efficiency(%EE), zeta potential, polydispersity index(PDI) and Inhibition concentration of 50% on MCF7 cell lines(IC50) were considered. A set of 16 trial were run and optimization was carried out. The factors showed significant effect on particle size, zeta potential and IC50 values. The optimized formulation combination was 2mg/ml chitosan concentration, 7mg/ml sodium TPP concentration, 7% acetic acid concentration and pH at 6. The responses were 630.92nm particle size, 0.685 PDI, percentage encapsulation efficiency of 97%, iC 50 at 136.85uM and -8.175 zeta potential. The desirability of the outcome was 0.861.
