Abstract

FORMULATION AND EVALUATION OF TRANSDERMAL DRUG DELIVERY SYSTEM OF METOCLOPRAMIDE HYDROCHLORIDE

ABSTRACT

The purpose of this research was to develop a matrix-type transdermal therapeutic system containing drug Metoclopramide hydrochloride using different ratios of ethyl cellulose and polyvinyl pyrrolidone polymers by solvent evaporation technique. Metoclopramide hydrochloride using different ratios of ethyl cellulose and polyvinyl pyrrolidone polymers by solvent evaporation technique. Different concentration of dibutyle phthatate was used as plasticizer and oleic acid was used as penetration enhancer. The result followed the released profile of Metoclopramide hydrochloride followed in-vivo drug release and ex-vivo permeation studies in different formulation. However, the release profile of the optimized formulation MH5 followed mixed zero-order and first order and indicate the permeation of the drug from the patches was governed by a diffusion mechanism. Ex-vivo permeation studies of formulation showed that the films containing higher proportion of polyvinyl pyrrolidone are suitable for once-a-day drug delivery and films containing higher proportion of ethyl cellulose showed sustained drug delivery through transdermal route for a period of more than 24 hrs.
