SOLUBILITY ENHANCEMENT OF ATORVASTATIN CALCIUM BY SOLID DISPERSION TECHNIQUE

ABSTRACT
The aim of present study was to enhance the solubility and dissolution rate of Atorvastatin calcium (ATC) by solid dispersion technique using polymer PEG 4000. Solid dispersions of ATC were formulated in different ratios by physical mixture and hot melt method 1:0.25, 1:0.5, 1:1, 1:2 and 1:3 using drug and polymer. The tablets were prepared by direct compression method. Tablets were physically characterized for various parameters like general appearance, weight variation, hardness, friability and evaluated for in vitro release characteristics for one hour in medium phosphate buffer at 37°c. The result showed that all the pre
-compression and post-compression parameter of tablets were within the limit of pharmacopeia. Among the two-method used, ATC solid dispersion prepared by hot melt method in 1:1 (f3) ratio showed most promising result in comparison to other ratios and physical mixture method.
