DEVELOPMENT AND EVALUATION OF GASTRO-RETENTIVE CEFADROXIL FLOATING MICROSPHERES

ABSTRACT
Cefadroxil is a broad spectrum first generation cephalosporin anti-biotic and it’s effective against a gram positive and gram negative bacteria, used for treatment of UTI and upper RTI. Present studies indicate that the development and evaluation of Cefadroxil gastro-retentive floating microspheres using Emulsion Solvent Evaporation Technique and Emulsion Gelation Technique with an aim to controlled release and improves the bioavailability. Six formulations were prepared with different concentration of ethyl cellulose, HPMC and sodium alginate. Depending upon the drug to polymer ratios, the percentage yield found between 47.60% to 69.08% and entrapment efficiency of floating
microspheres was 62.06% to 82.50% in all formulations. Prepared Microspheres showed a good specificity, spherical and uniform in shape with smooth surface and mean particle significantly increase with increasing polymer concentration. The percentage in-vitro buoyancy of floating microspheres was found in 64.24% to 84.42%. In-vitro drug release studies performed in simulated gastric fluid and revealed that, CEF 3 and CEF 6 formulation having 89.21% and 72.24% drug releases at end of dissolution studies. Stability studies of best formulation showed good results and there are no significant changes in drug content. In-vitro antibacterial activity of selected formulation exhibits better zone of Inhibition and confirming an antimicrobial activity. It could be conclude that all formulations were shown satisfactory results and suitable for potential therapeutic uses.
