COMPARATIVE ANALYSIS OF BENDAMUSTINE HYDROCHLORIDE CYTOTOXICITY IN HEALTHY HUMAN LYMPHOCYTES AND L5178Y TK+/- MOUSE LYMPHOMA CELLS

ABSTRACT
The study was conducted to evaluate and compare the cytotoxicity of
Bendamustine hydrochloride in Mammalian Chromosome Aberration
Test (CAT) In vitro, using cultured healthy human whole blood
lymphocytes and In vitro Cell Gene Mutation Assay (CGM) using
L5178Y TK+/- mouse lymphoma cells as per OECD guidelines No.
473 & 476 [1,2] respectively, both in the presence (2%v/v) and absence
of metabolic activation system up to the cytotoxicity. Bendamustine
hydrochloride is cytotoxic at the doses 300, 150, 75, 37.5, 18.75, 9.37
μg/ml of culture medium and non-cytotoxic at 4.68 μg/ml in CAT
where 300, 150, 75, 37.5μg/ml of doses are cytotoxic and 18.75, 9.37, 4.68μg/ml doses are
non-cytotoxic in CGM, both in the presence and absence of metabolic activation system.
Comparatively Bendamustine hydrochloride test doses are more cytotoxic in the presence of
metabolic activation system (2%v/v) than absence both in CGM, CAT and lower doses of
Bendamustine hydrochloride is cytotoxic in CAT when compare with CGM.
