PREPARATION AND EVALUATION OF VALSARTAN SOLID DISPERSION WITH HYDROPHILIC POLYMER (SOLUPLUS) BY FUSION METHOD

ABSTRACT
Valsartan is a potent, orally active nonpeptide tetrazole derivative and selectively inhibits Angiotensin Receptor type 1 which causes reduction in blood pressure and is used to treat hypertension. It is a poorly water soluble drug. Due to that its having poor bioavailability. The aim of the current study was to prepare solid dispersion of valsartan in order to enhance its solubility by using polymer like soluplus, pvp k-30 and poloxamer 407 which ultimately helps to increase bioavailability of valsartan. Solid dispersion of valsartan was prepared by using fusion method. The advantages of fusion method
are simple, cost effective, fastest. Solubility study of pure drug and physical mixture were carried out, and physical mixture prepared by using different carriers like valsartan: soluplus, valsartan: poloxamer 407, valsartan: soluplus: poloxamer 407, valsartan: soluplus: PVPK-30 out of this valsartan: soluplus (1:4) ratio shows more solubility. According to their solubility study data 1:4 ratio of valsartan: soluplus almost increases solubility. Solubility of the drug was increased by the fusion method as compared to pure drug. The bioavailability of solid dispersion formulation was also enhanced in comparison with Std. valsartan. Finally it can be concluded that, the solid dispersion technique is a useful technique in improving the solubility and bioavailability of slightly water-soluble compounds such as valsartan.
