Abstract

RP-HPLC METHOD DEVELOPMENT AND VALIDATION FOR SIMULTANEOUS ESTIMATION OF DIACEREIN AND GLUCOSAMINE SULPHATE IN TABLET DOSAGE FORM

ABSTRACT

A new, precise, rapid, accurate RP-HPLC method was developed for the Simultaneous Estimation of Diacerein and Glucosamine sulphate in tablet dosage form. After optimization the good chromatographic separation was achieved by Isocratic mode with a mixture of Acetonitrile: potassium dihydrgen phosphate Buffer pH 4.5 in the ratio of 50:50 v/v as the mobile phase with Zorbax C18 (4.6 x 250mm, 5m, Make: Agilent) column as stationary phase at flow rate of 0.8 mL/min and detection wavelength of 254 nm. The retention times for Diacerein and glucosamine sulphate found to be 1.9 and 3.8min respectively. The linearity of this method was found in the concentration range of 100μg/mL to 300 μg/mL for Diacerein and 1500 μg/mL to 4500 μg/mL for Glucosamine sulphate. The correlation coefficient R2 value is found to be 0.999 for Diacerein and 0.9998 for Glucosamine sulphate. The LOD and LOQ for Diacerein were found to be 2.715μg/mL and 9.050 μg/mL respectively. The LOD and LOQ for Glucosamine sulphate were found to be 2.8892μg/mL and 9.6308 μg/mL respectively. By this method percentage recovery for Diacerein and Glucosamine sulphate were found to be 100.00 and 100.00 respectively indicates that the proposed method is highly accurate.
