Abstract

MOLECULAR PHYLOGENY OF PTEROCARPUS MARSUPIUM AND PTEROCARPUS SANTALINUS

ABSTRACT

Pterocarpus santalinus and Pterocarpus marsupium are the endangered perennial species listed in the Red data book. Pterocarpus santalinus or Red Sanders is native to India, medicinally important in treating uterine bleeding and heartwood of the bark used to treat inflammation. Pterocarpus marsupium or the Indian Kino or the Malabar Kino Tree or the Kino Tree is a medium to large, deciduous tree native to India, Nepal, and Srilanka.This indicates the need for preservation and conservation efforts at the population level. In this randomly amplified polymorphic DNA (RAPD) technique was employed for the molecular characterization and Barcoding was used for molecular identification of Pterocarpus santalinus and Pterocarpus marsupium. Twenty two decamer oligonucleotide primers were used to amplify the genomic DNA which was isolated from the leaf samples. Out of twenty two primers used, five did not amplify, seven gave faint and non-reproducible bands, while ten gave species-specific reproducible unique bands. The unique bands obtained in PCR amplification clearly differentiated both the pterocarpus species from one another. Further study of genetic variability with rbcL markers gave the molecular taxonomical difference in these two Pterocarpus species.
