Abstract

DEVELOPMENT AND VALIDATION OF RP-HPLC METHOD FOR SIMULTANEOUS DETERMINATION OF PARACETAMOL, ACECLOFENAC SODIUM AND CHLORZOXAZONE IN COMBINED DOSAGE FROM

ABSTRACT

A simple, precise, accurate and rapid reverse phase high performance liquid chromatographic isocratic method has been developed for the estimation of Paracetamol (PCM), Aceclofenac (ACF), and Chlorzoxazone (CHZ) in tablet dosage form. The chromatographic separation was attained on WELCHROM C18 column (250 mm x 4.6 mm, 5μm) as stationary phase with mobile phase comprising phosphate buffer (pH-6.65): acetonitrile (60:40, v/v) with a flow rate of 1 ml/min, and UV detection at 280 nm. The retention times of PCM, ACF and CHZ were 3.203 min, 7.742 min, and 8.873 min respectively. The linearity of PCM, ACF and CHZ were in the range of 10-50 μg/mL, 2-10 μg/mL, and 5-25 μg/mL with correlation coefficient greater than 0.999. The mean % recovery of PCM, ACF and CHZ were found to be 100.24%, 100.20% and 100.16% respectively. The limit of detection (LOD) for PCM, ACF and CHZ was found to be 0.2749 μg/mL, 0.0759 μg/mL and 0.2891 μg/mL respectively and the limit of quantitation (LOQ) for PCM, ACF, and CHZ was found to be 0.8330 μg/mL, 0.2302 μg/mL and 0.8734 μg/mL respectively. The results of study show that the proposed RP- HPLC method was found to be simple, rapid, precise and accurate, which can be successfully used for the determination of PCM, ACF and CHZ in bulk drugs and in its pharmaceutical dosage forms.
