Abstract

STUDIES ON BACTERIAL SPECIES AND PHYSIOCHEMICAL PARAMETERS OF WATER FROM RIVER VARUNA

ABSTRACT

The aim of this study was to identify bacterial diversity in various sources of river Varuna (U.P.) and to determine their physiochemical parameters. The collected samples were processed for bacterial isolation using the selective media including Nutrient agar, Blood agar and MacConkey agar media. The conventional methods of spreading and streaking were used for isolation of bacteria from collected water samples. Pure colonies of isolates organisms were identified and characterized using standard gram staining and biochemical tests. The confirmation of the bacterial species was done by 16S rRNA amplification followed by the bioinformatic analysis of the sequence using alignment tool like BLAST. The bacteria were isolated from river water samples and it yielded some isolates representing 4 different types of bacterial species in our study i.e. E. coli, Salmonella spp., Shigella spp. and Streptococcus pyogenes. Varuna River Water samples were analyzed for physicochemical parameters including pH, BOD, COD, Acidity, Dissolved oxygen (DO), Chloride, Magnesium, Sodium, and Total hardness. In this study we focused on the Monsoon variations of physicochemical parameters in water samples from river Varuna of Varanasi. The purpose was to assess the quality of water from the sources in three different seasons (Pre- monsoon, Monsoon Post- monsoon). In this concluded that the water is commonly contaminated with different microbes and this infectivity may be playing a role in the show of potentially harmful organisms. It was concluded that clean water for all is one of the most challenges of the 21st century and that microbiological control of clean water should be the norm everywhere. Routine basic microbiological analysis of clean water should be carried out by assaying the presence of Escherichia coli and Streptococcus pyogenes by culture laboratory process.
