Abstract

ROXITHROMYCIN LOADED MICROSPHERES INCORPORATED IN IN-SITU GEL OF POLOXAMER 407 FOR PERIODONTITIS (RLMISG)

ABSTRACT

Aim of the present work was to develop a ideal system of roxithromycin loaded microspheres in in situ gel (RLMISG) as a treatment modality for periodontitis. The design consist of roxithromycin loaded microspheres which further incorporated in to in situ gel, in order to develop a controlled release drug delivery system of roxithromycin for periodontitis. The prepared roxithromycin loaded microspheres were evaluated for particle size, drug loading, entrapment efficiency and cumulative drug release. RLM I with optimal drug release (79.80±1.02 %) and particle size (89.32±0.02μm) was identified as the best formulation. RLMISG I showed optimal drug release (75.5±0.04%) upto168 hrs and less gelling time (29±0.62 sec) and temperature(29.78±0.43°C) in situ gelling syringeable periodontal sol of roxithromycin loaded microspheres was designed which is capable of controlled delivery of roxithromycin at periodontal cavity. The sol had palatability, Syringeability and inverted into gel at 34°C temperature. Thus in situ gelling controlled release syringeable periodontal drug delivery system with improved palatability, prolonged retention time in periodontal pocket and additive anti inflamatory potential was successfully formulated. Using such a device not only sustained an effective dose for the require length of time, but also has improve retention, ease of application and target localised area of periodontal cavity and more effective than the regular drug and medicines which act systemically. This device do not probe the risk over dose or systemic over load, simple for formulation.
