Abstract

BIO-DEGRADATION OF RED AZO DYE BY STREPTOMYCES CACAOI SUBSP. CACAOI .M20

ABSTRACT

There is lot of adverse effects like cancer and mutation in animals and human beings in the living systems were observed in the areas where the textile industries are located. Since the red azo dyes are released from textile mills, it should be degraded before it enters in to the living environment. An attempt was made to degrade the red azo dye using 25 mangrove actinomycetes isolated from soil sample of Avicennia marina. It was noted that the isolate M20 was the only strain that degraded the red azo dye. To study the bioaccumulation and degradation of red azo dyes by the isolate M20 - Streptomyces cacaoi subsp. cacaoi, initially, 60 ml of modified nutrient glucose agar was sterilized with 0.080g/100ml of red azo dye, dispensed into required number of plates allowed to solidify. The plates were streak inoculated in the centre with isolate M20 and incubated at the 30ºC for 7 days. Then the study of bioaccumulation of red azo dye was further examined by inoculating the loop of inoculam of isolate M20 in the test tubes that contain each sterilized 12.5 ml of modified nutrient glucose broth with 0.005 g, 0.010 g, 0.020 g of red azo dyes and incubated at the 30ºC for 15 days. Discolouration of red azo dye by the isolate M20 was observed 4th onwards in the liquid media. Discolouration of red azo dye by the isolate M20 was assessed in 10thday for degradation using ultra violet- visible spectrophotometer.
