Abstract

FORMULATION AND EVALUATION OF SUSTAINED RELEASE MATRIX TABLETS OF METFORMIN BY USING THE COMBINATION OF NATURAL AND SYNTHETIC POLYMERS

ABSTRACT

The main objective of the present work was to develop sustained release matrix tablets of Metformin using gum olibanum alone and in combination with HPMC and Na.CMC. Tablets were prepared by the wet granulation method. The physicochemical characteristics of polymer (Gum olibanum), and pre-compressional parameters and postcompressional parameters were evaluated, all the parameters were found to be within the limits. The dissolution studies were performed using USP apparatus type-2 using phosphate bufferpH 6.8 as dissolution medium. The drug release was dependent on the type and concentration of the polymer. The results of studies showed that the gum olibanum alone is not able to control the drug release efficiently, however the combination of gum olibanum with other polymers (HPMC, Na.CMC) could efficiently produce the sustaining effect. Among all the formulations of metformin, the gum olibanum in combination with HPMC which is (F9) was found to possess better controlled release of metformin over a period of 12hrs. The optimized formulation also subjected to different Kinetic models and it was shown that the formulation follows Higuchi model and from that it was concluded that the rate controlling mechanism is Diffusion. From the KorsmeyerPeppas equation it was found that drug release follows super case II transport. The final optimized batch was kept for 3 months of Accelerated stability study according toICH guidelines and formulation was found to be stable after 3 months of study.
