Abstract

EFFECT OF WHEAT GRASS EXTRACTS (TRITICUM AESTIVUM) ON WOUND HEALING RELATED PROTEINS (SYNDECAN4 AND TISSUE TRANSGLUTAMINASE)

ABSTRACT

Improper healing of the wound is still the problem in the surgery. Diabetes, obesity, immune deficiency & protein deficiencies all play a role in this improper healing. In medical terminology, wound can be defined as, an injury, where the skin, is either cut, torn or punctured. This wound ultimately leads to the damage of the dermis, and thereby destructs the structural integrity of the skin. The experiment is designed to investigate for the first time to study the beneficial role of the wheat grass extracts on the fibroblast cell lines and to trace out its effect on the TG2 and syndecan4 proteins. The study was designed to isolate the phenolic compounds from the wheat grass extracts and to check for the expression of TG2 and Syndecan4 in the fibroblast cell lines with and without treatment (extract). The study was also aimed to scan the levels of TG2 expression on the surface of the cells using Confocal fluorescence microscopy. The syndecan4 gene was 304 times more expressed than Control at 100μg/ml, and syndecan4 was 104.1 times more expressed than control at 200μg/ml. Similarly in case of the TG2 expression. TG2 was 174.85 times more expressed than control at 100μg/ml and 118.32 times more expressed than control at 200μg/ml. The extract showed positive response towards the TG2 expression on the cell surface. The activity of the extract on the TG2 expression was confirmed with the insitu activity. Further the experiment need to be planned for optimization of the concentration of the extract and its cytotoxicity.
