Abstract

DESIGN OF A LOW COST FERMENTATION MEDIUM FOR THE PRODUCTION OF SERRATIOPEPTIDASE ENZYME BY A NOVEL Streptomyces sp

ABSTRACT

Horse gram (Microtylona uniflorum) is a novel substrate for the production of serratiopeptidase by using a novel source Streptomyces hydrogenans var. under solid state fermentation, was first recognized in the present study. Various bioprocess parameters, were optimized by one factor at a time approach manner. The Maximum yield 85 U/gds was achieved with the following optimized fermentation parameters: Solid substrate (Horse gram)), particle size (<250 micron), substrate concentration (5gms), inoculum concentration (20%v/w), initial moisture content (40 %v/w) and additional nitrogen source (Soyabean meal (1%). It was observed that pH strongly influences the serratiopeptidase production by Streptomyces hydrogenans, the ions present in the buffer decreases the growth of micro organism, there by reduced the enzyme production. The maximum yield of serratiopeptidase was recorded with control (horse gram production medium pH-6.5-7). It was also observed that additional mineral salts, carbon sources in the medium was not showed any significant enzyme production. These results indicate that the protein, carbohydrate and iron which are naturally present in horse gram were sufficient for the growth of Streptomyces hydrogenans var. to produce a miracle enzyme serratiopeptidase. As the horse gram is not only a low cost pulse with rich protein content but also have some medicinal values like dissolving kidney stones and reducing high blood pressure.[16] so the present findings opened new vistas in the utilization of horse gram as a substrate for solid state fermentation.
