Abstract

ANALYSIS OF ACETYLCHOLINESTERASE ACTIVITY FROM PLANT EXTRACTS USING GC-MS AND AUTODOCK

Kiran Kumar S. J.* and Shwetha R. Gowda

ABSTRACT

Acetylcholine esterase (AchE) inhibitors have therapeutic applications in Alzheimers disease. The brain of a freshly sacrificed zebra fish was used as a source of Acetylcholine esterase. Plants belonging to families Acanthaceae, Apocynaceae, Amaryllidaceae, leguminosae, lamiaceae have been reported with acetylcholine esterase which has an inhibitory potential. Bioassay directed phytochemical investigation on Crinum asiaticum have led to the isolation of AchE inhibitors and GC-MS analysis gave 19 compounds from the methanolic extract. Four out of 19 compounds found in GC MS analysis were selected based on their acidic property and availability of their structures were used for enzyme- ligand interaction with the programme Autodock 1.1.2
version were the compounds docks to receptor. The results clearly indicates the wide possibility of this plant to be used in the treatment of Alzheimer’s disease.

Keywords: Acanthaceae, Apocynaceae, Amaryllidaceae, leguminosae, lamiaceae.
