Abstract

EVALUATION OF NEW OXCARBAZEPINE ANALOGUES FOR DIABETIC NEUROPATHY PAIN

ABSTRACT

Neuropathic pain in diabetes is a challenge for health care industry. However many drugs are available such as anticonvulsants, tricyclic antidepressants to treat the above condition, but their effect is partial .Because of the limitation of the available treatment, new agents that are effective and safe should be explore and investigate through research for diabetic neuropathy treatment. Oxcarbazepine is a relatively novel anticonvulsant drug derived form Carbamazepine. It has fewer side effects and less drug -drug interaction then carbamazepine, because it is metabolised by non –CYP450 pathway. To potentiate the activity of oxcarbazepine for diabetic neuropathy six analogues were synthesized by changing the keto group of 10th position and NH2 group of 5th position with different groups. Then synthesized oxcarbazepine analogues were screened for diabetic neuropathy pain. STZ 50mg/kg was used to induce diabetes in rats in control and testing groups. The six synthesized analogues were given in the dose of 50mg/kg, then evaluation of the drugs done by hot plate and Randall Selitto test respectively. Among the six analogues, four were shown activity against diabetic neuropathy pain where as remaining two were not significant.
