Abstract

FORMULATION OF ORAL PULSATILE TABLET OF GLIPIZIDE BY PRESS COATING TECHNIQUE AND ITS IN-VITRO EVALUATION

Rohit Shewale*, Akshay Thorat, Rajesh Jagtap and Rajendra Doijad

ABSTRACT

The aim of present investigation deals with the formulation and evaluation of oral pulsatile drug delivery system (PDDS) for antidiabetic drug. The basic idea behind the dosage form development is to investigate effect of coating polymer conc. on lag time and drug release from directly compressed tablet. Present study also aimed to develop a system that would reduce dosing frequency and improve patient’s compliance. All the ingredients and polymers such as Guar gum, HPMC K100M, Cross-carmalose sodium, microcrystalline cellulose, Talc, Magnesium stearate were procured from Loba chemicals Mumbai. Glipizide was received as a gift sample from Lupin Pharmaceuticals Ltd. Pune. The method applied for the preparation of tablet was direct compression method. The results of in-vitro drug release studies showed that formulation (API and Guar gum and HPMC K-100) has better drug release after 6 hrs. lag time. Glipizide direct compression tablet for time controlled oral pulsatile drug delivery system was successfully developed and prepared by using the natural polymers in different concentration and evaluated for pre-compression, post compression parameters, in-vitro drug release studies and stability studies. It was observed that the increasing or decreasing concentration of polymer blend had affected on the drug release. As we increases the concentrations of polymer blend the lag time will be increases & vice versa.

Keywords: Pulsatile drug delivery system (PDDS), Lag time, Glipizide, and Drug release.
