Abstract

DEVELOPMENT AND EVALUATION OF RIZATRIPTAN PATCHES FOR BUCCAL

ABSTRACT

Rizatriptan is an Anti-migraine & Anti-Inflammatory Agent used in the treatment of acute migraine attacks. After oral administration of tablet it undergo metabolism and resulting in to the inactive indole acetic acid derivative in turn which results into the decreased bioavailability (45%) of rizatriptan. So, in order to improve the bioavailability and efficacy of rizatriptan, we have prepared buccal patches of rizatriptan. With the preparation of buccal patches we are delivering the drug through buccal route by which we can bypass the presystemic metabolism of the rizatriptan. In the present piece of research work, Rizatriptan Buccal Patches were prepared using mucoadhesive polymers like HPMC, Chitosan, NaCMC & HEC by Solvent Casting technique. Buccal patches were characterized for number of parameters like physical appearance & surface texture, weight uniformity, thickness, folding endurance, swelling index, bursting strength, Surface pH, drug–Excipient interaction study, drug content uniformity and In vitro drug release study. Drug content uniformity study showed uniform dispersion of the drug throughout the formulation in the range of 96.477 ± 1.385 to 99.460 ± 0.147 %. Invitro drug release studies showed that more than 90% of the drug was released at the end of 10th hrs.
