Abstract

FORMULATION DEVELOPMENT AND EVALUATION OF THERMOSENSITIVE ORAL GEL CONTAINING DOXYCYCLINE HYCLATE

ABSTRACT

The objective of this research was to formulate and evaluate thermosensitive oral gel containing Doxycycline hyclate in combination with poloxamer 188 and carbopol 934. The thermosensitive oral gel containing Doxycycline hyclate was prepared by cold method using poloxamer 188 as thermosensitive gel and carbopol 934 as mucoadhesive polymer. All the prepared formulations were evaluated for different parameters including study of the visual appearance, determination of the pH, gelation and gel melting temperature, viscosity gel strength, spreadability and mucoadhesive strength.The pH of the formulation was adjusted within the range of 6.83±0.07 - 7.09±0.29 which is compatible with the pH of the oral cavity. Gelation temperature was found to be within 32±0.26 –39±0.20°C. The gel melting temperature was found to be in the range of 74± 0.13 - 80±0.18. The increase in temperature increases the viscosity of the formulations which in turn decreases the spreadability. Gel strength was found to be higher as the viscosity increases. Drug content was within the range of 92±0.13% - 97±0.09% which indicates the uniformity of content. The increase in concentration of carbopol 934 increases the mucoadhesivity of the formulations.In this research work the results obtained after the evaluations of the formulations indicate that the evaluated values of Formulations F5-F10 were found to be ideal for administration into the oral mucosa.
