Abstract

ZEBRAFISH AS A MODEL FOR HYPERPIGMENTATION USING MANGIFERA INDICA

ABSTRACT

The enzyme tyrosine ammonia lyase (TAL) present in most of the plants has the ability to breakdown the tyrosine to p-coumaric acid and elimination of non oxidative ammonia. We extracted the crude enzyme from mangifera indica; presence of TAL was confirmed by the formation of p-coumaric acid was detected using UV-VIS spectrophotometer. To enhance the enzyme activity both metal ions and organic compounds extracted from plant phyllanthus emblica were used. The compounds were identified by using gc-ms.All the compounds were docked to TAL structure using Autodock software; cholesta-5, 22-dien-3-ol was identified with high affinity. Then the embryos of zebrafish were used to study enzyme activity in vivo condition. TAL enzyme was treated with embryos before the pigmentation on the skin with control where enzyme was not added. TAL treated embryos has not shown pigmentation on skin where control shows the pigmentations.
